I make characters on a Game engine called Mugen. I am considering making an Ender's Game 2d battle game. (Like street fighter.)
but then I reconsidered it....I am more so considering making an entire Orson Scott Card verse game. what do you guys think?
1. Who would the main 10 characters be? 2. Who would the bosses be. 3. What would the moves for the enders character games be like? I mean they are just humans. Which is why I think that using his whole universe would be a better. A few characters from each.

I think this is a very bad idea, as Chair Entertainment would sue you out of existence if you actually released it.

You don't have to sell anything to be guilty of copyright or trademark infringement. You're probably going to be in LESS danger of being sued successfully for damages if you don't sell anything, but you could still be guilty of infringement.
I think another way to look at this is as a question of politeness. Based on what I've seen Card write in the past, I think he would disapprove (strongly) of an unauthorized derivative use of his intellectual property. Therefore it would be impolite to proceed without permission.

Aww... This dissapoints me. I was dissapointed when I read that chair got the rights to make the Enders Game game. I was hoping to hava part in making a game like that (in fact, this is why I joined the forum inthe first place. I was going to use DarkBASIC and FPSC to make an RPG where you play as a battle schooler and can earn the roles of Ender, Bean, or make your own destiny.

Agreed... Also, what kind of company calls themselves Chair Entertainment? Its like, ok boys, what shall we call ourselves? Hmmmm.... Oh look! A chair! Hmmmm.... CHAIR ENTERTAINMENT! That will totally make everyone want to get off their. . . chairs. . . huh? HUH!? Am I right! And go down to the gamestore and pick up a Chair Entertainment game! Boom Bam! SELL IT!

I'd have the same advice even if there were no video games, movies, t-shirts, or bobbleheads. The enderverse was never *not* commercialized. It exists in stories that were written for profit.
