If my book has an enormously important twist ending, do I reveal that to any agents I query? What if it's something I would NOT put on the backcover, for fear of spoiling the surprise?

You usually do not reveal the ending in your query. In fact, there are agents (Kristin Nelson, for example) who say that you don't need to go further than the inciting incident in the query.

You do reveal the ending in the synopsis, though.

Agreed. Queries are teasers to get the agent interested. Synposes are proof yuo have a complete coherent plot.
